The clue is in the article un/una or el/la.
Where is there a florist's? ( = Where can I find a florist's?
¿Dónde hay una floristería?
vs
Where is the florists?
¿Dónde está la floristería?
On the first one ¿Dónde hay una floristería? you're asking whether one exists nearby, not about a specific one.
On the second one ¿Dónde está la floristería? you're referring to a specific florist that both speakers know or have already mentioned.
